增强型皮克级化学发光液是一种采用新型配方的辣根过氧化物酶(HRP)底物化学发光液,具有灵敏度极高、性能稳定、半衰期长、背景(本底)极低、重复性好等优点。它可以用于固相膜(如 NC、PVDF)上结合的蛋白(Western Blot)或核酸(Southern/Northern Blot)的检测,既可感光X光胶片,也适用于荧光CCD相机的荧光扫描。
高灵敏度
可检测低至1pg~100fg的抗原,适用于痕量抗原的检测本底极低
即使长时间曝光,背景仍然极低,远远优于国产与进口同类产品半衰期长
信号衰减慢,6h以上仍可感光X光胶片稳定性好
溶液可在室温避光保存12个月转膜及抗体结合。采用常规方法进行转膜、封闭、一抗、HRP-二抗结合,直至二抗洗涤完毕;
等体积混匀P-ECL A和P-ECL B,配制化学发光工作液(每平方厘米的转印膜约需0.1mL的工作液);
将化学发光液均匀滴到膜上,反应约3min。如要检测的蛋白信号很高时,请尽量不要让化学发光工作液处于静止状态,可将加了工作液的膜置于摇床上混动约3min,以避免局部的反应物过度消耗,影响检测信号的线性范围;
取出膜,沥去多余的发光工作液,将膜包裹于透明的保护夹中;
X光片曝光:在暗室中将膜对X光片曝光。曝光时间从几秒到几分钟不等。将底片显影,如有必要,可根据显影结果调整曝光时间进行第二次曝光;
CCD成像仪:无需将膜置于保护夹中,可直接用于拍照。拍照按仪器使用说明进行;
使用过的膜可剥离抗体后进行多次杂交。
1. P-ECL A及发光工作液应避免强光照射而导致灵敏度降低;
2. 增强型皮克级化学发光液灵敏度很高,因此应避免由于使用的抗体浓度过高而造成背景升高现象;
3. NaN3会抑制HRP的活性,因此二抗的保存或回收应避免使用NaN3;
