本品是一种经过改良的以溴酚蓝为染料,5倍浓缩的蛋白上样缓冲液。本产品中含有变性试剂,但是不含DTT、巯基乙醇等还原剂。可以用于常规的变性非还原型聚丙烯酰胺凝胶电泳的蛋白样品上样。
1.使用时,在蛋白质样品中加入1/4体积本产品,混匀;
2.将混合物用沸水浴加热3~5min;
3.待冷却后,高速离心5min,上清即可用于电泳分析。如蛋白样品在加入上样缓冲液后,无不溶性残渣,则只需短暂离心,将混合液收集到管底即可。加热后,如果管盖上出现较多冷凝水,建议轻弹离心管将冷凝水和样品混合以后,再进行高速离心步骤,此举有助于降低上样样品浓度不均的现象。
